i had biochemistry test the other day, and there was a question like this….?
"what type of purification did you see with the recombinant protein?".
what is this question asking? types of chromatography used, like Ni affinity chromatography???
how could i answer this question?

Not having seen the Q in its entirety I surmise that it is referring to Ni chelate isolation of Histidine tagged proteins. Nickel forms square planar complexes with electron donating groups or atoms such as the electron pair on the nitrogen in the imidazole group of histidine. So that would be the best bet given the syntax.


Short answer – Ni affinity chromatography could be correct.
Purification is simply isolating what you are interested in (in this case – the recombinant protein), so any technique that isolates is a possibility. For protein, metal affinity chromatography is a common method to use, so if you saw that in your lab – it would make sense to be the answer.
Of course, without being there and seeing all the techniques shown to you, I cannot be sure – but it makes sense as an appropriate answer.
